🧬 Complement System Activation: The Three Pathways Explained
The complement system is a crucial part of the innate immune system, composed of over 30 proteins that work together to fight infections, clear damaged cells, and promote inflammation. It can be activated through three distinct pathways, each triggered by different signals but all converging on a common goal: the destruction of pathogens.
1️⃣ Classical Pathway
Trigger: Activated by antibodies (IgG or IgM) bound to antigens on the surface of pathogens.
Mechanism: The binding of antibody-antigen complexes recruits the protein C1 complex (C1q, C1r, C1s), which then activates C2 and C4.
Outcome: Formation of the C3 convertase (C4b2a), leading to cleavage of C3 and amplification of the immune response.
Key Role: Bridges adaptive immunity (antibody-based) and innate defense.
2️⃣ Lectin Pathway
Trigger: Activated by mannose-binding lectin (MBL) or ficolins binding to specific sugars (like mannose) on microbial surfaces.
Mechanism: Similar to the classical pathway, but initiated without antibodies. MBL-associated serine proteases (MASPs) cleave C4 and C2 to form the C3 convertase (C4b2a).
Outcome: Activation proceeds similarly to the classical pathway.
Key Role: Provides an antibody-independent defense, especially early in infection.
3️⃣ Alternative Pathway
Trigger: Spontaneous hydrolysis of C3 in the plasma (a "tick-over" mechanism), or direct recognition of pathogen surfaces.
Mechanism: Hydrolyzed C3 binds to factor B, which is then cleaved by factor D, forming C3bBb, the alternative pathway's C3 convertase.
Outcome: Rapid amplification of C3 cleavage and complement activation, especially on microbial surfaces.
Key Role: Provides a constant surveillance mechanism and amplifies the effects of the other two pathways.

Opsonization: Marking microbes for destruction
Inflammation: Attracting immune cells to infection sites
Membrane Attack Complex (MAC) formation: Directly lysing and killing pathogens
